Protective effect of sucralfate against alcohol-induced gastric mucosal injury in the rat. Macroscopic, histologic, ultrastructural, and functional time sequence analysis.

摘要: Histologic or ultrastructural evidence of the ability sucralfate to protect gastric mucosa against ethanol injury is lacking. Therefore we analyzed morphologic and functional changes in 120 rats receiving, intragastrically, 2 ml either 500 mg/kg body wt a control solution 1 h later 100% ethanol. At 15 min, 1, 4, 6, 24 after instillation, mucosal were assessed by macroscopic examination, quantitative histology, scanning electron microscopy, recordings potential difference, measurements volume, pH, electrolytes contents. Between min necrotic lesions controls involved greater than 33% area sucralfate-treated group less 4% (p 0.001 for each period). In controls, instillation produced surface epithelial cell disruption deep (greater 0.2 mm) necrosis involving 55% +/- 3% length. sucralfate-pretreated animals, epithelium was present at h, 4 but virtually absent (0%-2%; p vs. controls) during entire study period. The mostly reestablished 6 not controls. We concluded that protects ethanol-induced preventing as consequence proliferative zone cells rapidly restitute integrity.
