Transforming growth factor-beta and interleukin-1 modulate metalloproteinase expression by corneal stromal cells.

摘要: The enzyme collagenase participates in remodeling the extracellular matrix of corneal stroma during normal wound healing and mediates degradation that occurs many pathologic states. Because this is synthesized secreted by cells, therapy degradative disorders might be geared toward control expression. effects two cytokines, transforming growth factor-beta (TGF-beta) interleukin-1 (IL-1), on expression cultured stromal cells are reported. In addition, concomitant these cytokines three additional members metalloproteinase (MMP) family--stromelysin, 72-kilodalton (kD) gelatinase, 92-kD gelatinase--were investigated. When situated stroma, they produce only a single MMP, 72-kD gelatinase. This pattern was reproduced freshly plated primary culture. However after passage culture, also began to secrete stromelysin. Treatment cultures with recombinant human IL-1 induced stromelysin gelatinase increased further treatment. passaged or IL-1-treated TGF-beta reverted exhibited primary, untreated cultures, ie, repressed while increased. These results suggest may important agents for controlling MMP healthy diseased corneas.
