摘要: We have studied the synthesis and expression of surface proteins in zygotes Plasmodium gallinaceum during their transformation to mature ookinetes. The cells were biosynthetically labelled vitro using [35S]methionine immunoprecipitated with rabbit anti-ookinete serum or monoclonal antibodies. Early (approx. 2 h post-gametogenesis fertilization) synthesized expressed on a protein Mr 26 000 as observed under reducing conditions polyacrylamide gel electrophoresis presence sodium dodecyl sulfate (SDS-PAGE) (31 non-reducing conditions) continued do so for 8–10 h; thereafter declined little none was ookinetes (>20 post-gametogenesis). Between 3–5 post-gametogenesis, also began synthesize 28 (34 conditions). Synthesis this throughout development; predominant by ookinete. been designated earlier PgO-1 PgO-2 respectively (Carter Kaushal, Mol. Biochem. Parasitol. (1984) 13, 235–241). Neither gametocytes prior gametogenesis. Both could be [3H]glucosamine [3H]mannose. When incubated [3H]palmitic acid both bound fatty acids covalent linkage. two not otherwise appear structurally related. They differentially different antibodies gave rise distinct patterns peptides following digestion proteases such Staphylococcus aureus V-8, trypsin chymotrypsin.
