Analysis of cerbB2 expression using a panel of 6 commercially available antibodies

摘要: Summary Results are presented of a study comparing cerbB2 (neu or Her2) expression as assessed immunohistochemically in breast neoplasia using panel 6 commercially available antibodies. The antibodies were examined utilizing conventional formalin fixed paraffin embedded tissue, and compared with molecular analysis gene amplification. aim was to determine the practical utility each antibody, assessing ease use, specific non-specific staining characteristics, expense, thus allowing recommendation antibody choice for immunohistochemical assessment expression. Reassuringly, amongst 38 lesions (36 carcinomas, 2 fibroadenomas) subjected immunohistochemistry (IHC) (Ab), no gross discrepancy pattern seen. Of cases, 10 positive (26%), where at least one Ab demonstrated clear cytoplasmic membrane staining. total 45 (43 fibroadenomas), including all those by IHC, number cases showing amplification DNA 14 (31%). Using base line comparison, (Ne. 4) found stain 6’ carcinoma that level. Four Abs (1,3,5,6) stained 5 these cases. However, 3,4 displayed artefactual (in absence staining) precluded use reagents. Therefore, based on additional considerations cost No. 1 finally chosen from panel.
