(Z)-5-(2,4-Dihydroxybenzylidene)thiazolidine-2,4-dione Prevents UVB-Induced Melanogenesis and Wrinkle Formation through Suppressing Oxidative Stress in HRM-2 Hairless Mice

摘要: Background. Uncontrolled melanogenesis and wrinkle formation are an indication of photoaging. Our previous studies demonstrated that (Z)-5-(2,4-dihydroxybenzylidene)thiazolidine-2,4-dione (MHY498) inhibited tyrosinase activity in vitro. Objective. To examine vivo effects MHY498 as antiaging compound on UVB-induced formation, we topically applied dorsal skin HRM-2 hairless mice. Methods. Using histological analysis, evaluated after UVB exposure. In addition, related molecular signaling pathways were examined using western blotting, fluorometric assay, enzyme-linked immunosorbent assay. Results. suppressed by inhibiting phosphorylation CREB translocation MITF protein into the nucleus, which key factors for expression. Consistently, levels notably reduced mice treatment. Furthermore, collagen fiber destruction increasing type 1 procollagen concentration decreasing expression MMPs, play essential role degradation. As a mechanism, ameliorated oxidative stress NF-κB activation dermal Conclusion. study suggests can be used therapeutic or cosmetic agent preventing uncontrolled formation.
