摘要: Abstract Because tyrosinase catalyzes essential steps in melanin synthesis, inhibitors are widely used for skin–lightening. To reduce the side effects of synthetic compounds, safer isorated from natural products have gained attention. Here, we examined anti-melanogenic effect seed Ziziphus jujuba and identified eight major which spinosin exhibited strongest inhibitory activity (IC50 = 47 µM). When further examined, suppressed αMSH- or UVB-induced melanogenesis B16F10 cells without cytotoxicity. The was also shown a human skin model. As an underlying mechanism, silico analysis showed that may bind to suppress by forming multiple hydrogen bonds hydrophobic interactions with binding pocket tyrosinase. Lineweaver-Burk Dixon plots revealed acts as competitive inhibitor (Vmax, 11.61 × 103, 11.63 × 103, 11.62 × 103, Km, 0.139, 0.185, 0.239 con, 10 µM, 50 µM spinosin, respectively). In conclusion, compound jujuba, could be novel additive treating pigmentation disorders skin-lightening cosmetics.
