Pirt, a TRPV1 Modulator, Is Required for Histamine-Dependent and -Independent Itch

摘要: Itch, or pruritus, is an important clinical problem whose molecular basis has yet to be understood. Recent work begun identify genes that contribute detecting itch at the level. Here we show Pirt, known play a vital part in sensing pain through modulation of transient receptor potential vanilloid 1 (TRPV1) channel, also necessary for proper sensation. Pirt−/− mice exhibit deficits cellular and behavioral responses various itch-inducing compounds, pruritogens. Pirt contributes both histaminergic nonhistaminergic and, crucially, involved forms are TRPV1-dependent -independent. Our findings demonstrate function extends beyond nociception via TRPV1 regulation its role as critical component several signaling pathways.
