摘要: We recently described culture conditions that allow proliferation of metastatic human breast cancer cells from biopsy specimens certain patient samples. These resulted in the development an immortalized cell strain designated SUM-44PE. same were used to isolate a lymph node separate patient. The SUM-16LN isolated this specimen cultured either serum-free medium or serum-containing supplemented with insulin and hydrocortisone. Unlike SUM-44PE have proliferated continuously for over two years, approximately 200 days underwent 15 20 population doublings before undergoing senescence. No beyond passage 8. keratin-19 positive had aneuploid karyotype. overexpressed p53 protein amplified epidermal growth factor (EGF) receptor gene high level expression tyrosine phosphorylated EGF protein. Despite presence levels these cells, they serum-free, EGF-free did not secrete detectable EGF-like mitogenic factor. In addition, potently inhibited by all concentrations exogenous tested neutralizing antibody Mab 425. results suggest present is direct result overexpression simulatory ligand.
