摘要: Previous studies have demonstrated that the administration of epidermal growth factor (EGF) to neonatal mice results in tyrosine phosphorylation multiple substrates all organs examined (Donaldson, R. W., and Cohen, S. (1992) Proc. Natl. Acad. Sci. U.S.A. 89, 8477-8481). One these substrates, a 55-kDa protein, was shown associate with receptor for EGF (EGFR). We now report identification this receptor-associated protein as SHC. Immunoprecipitation Western blotting analyses revealed SHC associates only activated EGFR. In absence stimulation, exists liver cytoplasmic monomer. Intraperitoneal injection results, within minutes, translocation 50-80% plasma membrane. The membrane-associated found be tyrosine-phosphorylated; subsequent release from membrane correlated dephosphorylation. conclude is physiological substrate appears participate vivo signaling response EGF.
