The protective effect of vildagliptin in chronic experimental cyclosporine A-induced hepatotoxicity

摘要: The study examined the effect of dipeptidyl peptidase-4 (DPP-4) inhibitor, vildagliptin, in cyclosporine (CsA)-induced hepatotoxicity. Rats were divided into 4 groups treated for 28 days: control (vehicle), vildagliptin (10 mg/kg, orally), CsA (20 s.c.), and CsA-vildagliptin group. Liver function was assessed by measuring serum levels aspartate aminotransferase (AST), alanine (ALT), gamma glutamyltransferase (γGT), lactate dehydrogenase (LDH), albumin, histopathological changes liver examined. Oxidative stress markers evaluated. Assessment nuclear factor-kappa B (NF-κB) activity hepatic extract, DPP-4, expression Bax Bcl2 also done. CsA-induced hepatotoxicity evidenced increase AST, ALT, γGT; a decrease albumin; significant alteration architecture. Also, thiobarbituric acid reactive substance (TBARS) superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), (GSH) levels, increased proteins with deceased Bcl2, NF-κB DPP-4 level observed upon treatment. Vildagliptin significantly improved all altered parameters induced administration. has potential to protect against reducing oxidative stress, activity, apoptosis, inflammation.
