摘要: Stimulation of the macrophage-like cell line THP-1 with phorbol ester 12-myristate 13-acetate (PMA) resulted in differentiation into cells many features macrophages. This was accompanied by transcriptional activation lipoprotein lipase (LPL) and apo E genes accumulation their protein products media. PMA-induced HEL HL-60 lines not induction gene for LPL, whereas induced slightly cells. By contrast, superoxide dismutase (SOD-1) either unaffected (THP-1) or down regulated (HL-60 cells) PMA treatment. Induction LPL mRNA dependent upon concentration added. A minimal 1.6 x 10(-8) M necessary macrophage differentiation, mRNA, synthesis enzyme. accumulates within 3 h after stimulation attains a maximum 6 h, thereafter slowly decreasing over next days. In steady-state level same cultures increased gradually period 48 induction. These studies thus demonstrate that are value as model to study quantitative temporal expression during differentiation.
