A surgical model of composite cystoplasty with cultured urothelial cells: a controlled study of gross outcome and urothelial phenotype.

摘要: OBJECTIVES To study the outcome of composite cystoplasty using cultured urothelial cells combined with de-epithelialized colon or uterus in a porcine surgical model, appropriate controls, and to characterize neo-epithelium created by cystoplasty. MATERIALS AND METHODS Urothelial were isolated propagated vitro from open bladder biopsies taken nine female minipigs. Cohesive sheets confluent transferred polyglactin carrier meshes sutured autologous four animals five. These segments then used for augmentation cystoplasty. Conventional colocystoplasty, colocystoplasty sham operations carried out six control animals. After killing at ≈ 90 days bladders removed examination immunohistochemical analysis, panel antibodies against cytokeratins differentiation-associated antigens. RESULTS Macroscopically, augmented derived uterine muscle had no evidence shrinkage contracture. Histological analysis showed that five uterocystoplasties, neo-urothelium was stratified transitional morphology, although some areas coverage incomplete. Immunohistochemical squamous differentiation both native segments. All colonic significant contraction poor coverage, reflecting unsuitability thin-walled de-epithelialization.
