31P NMR analysis of the DNA conformation induced by protein binding SRY/DNA complexes.

摘要: Complexes of the HMG box protein SRY with two duplexes 8 and 14 base pairs have been studied by 31P NMR complete assignment all phosphorus signals bound DNA are presented. While for free DNA, display limited spectral dispersion (< 0.8 p.p.m.) duplexes, resonances spread over 2 p.p.m. Based on previously published 3D structure hSRY-HMG, 8 mer it is demonstrated that upfield shifted correspond to site partial intercalation an isoleucine side chain into DNA. Moreover, observation significant difference in linewidths between allows estimate lifetime complexes from 31P–31P 2D exchange experiments.
