摘要: Rational strategies for the development of novel antidepressant agents are based on an analysis perceived therapeutic needs, which in turn derive from inadequacies therapies that currently available. The traditional (first-generation) antidepressants suffer three major drawbacks: They have unacceptable side effects; their efficacy is low; and effects develop slowly, typically requiring 3–4 wk treatment before clear improvements mood apparent. Since 1970s, these problems formed targets drug development, since any newly developed agent could only be marketed basis improvement over one or more areas.
