摘要: TCR-gamma delta cells, a T cell subset present in the epithelial and lymphoid tissues, have been implicated viral bacterial infections. We identified clone (TgI4.4) that, unlike TCR-alpha beta recognizes herpes simplex virus type 1 transmembrane glycoprotein, gI, an MHC class I- II-independent fashion. The TCR of TgI4.4 is composed rearranged V 8 (a alpha 2 family member) gamma 1.2 variable genes, heterodimeric pair not previously described. Furthermore, anti-V mAbs are sufficient to block recognition gI ligand. Strikingly, anti-gI Abs also capable blocking recognition, phenomena that very rare Ag recognition. Therefore, dissect mechanism involved this unique form we constructed mutant gIt, lacks surface expression upon transfection into APCs. This was for presentation. In contrast, wild-type expressed Ag-processing cell, RMA-S, recognized by TgI4.4, suggesting presentation occurs independently classical pathways. fact, through use soluble recombinant molecule, gI-Ig, show can recognize whole, unprocessed protein absence any These results suggest there exist alternate novel forms clone, may represent during pathogenic challenge, respond directly Ags on surfaces bacteria viruses.
