摘要: A miniature 96 sample ELISA-lab-on-a-chip (ELISA-LOC) was designed, fabricated, and tested for immunological detection of Staphylococcal Enterotoxin B (SEB). The chip integrates a simple microfluidics system into ninety-six plate, allowing the user to carry out an assay without laboratory. Assay reagents are delivered plate need separate devices commonly used in immunoassays. ELISA-LOC constructed using Laminated Object Manufacturing (LOM) technology assemble six layers with acrylic (poly(methyl methacrylate) (PMMA)) core five polycarbonate micromachined by CO(2) laser. has three main functional elements: reagent loading fluidics, wells, removal "surface tension" valve control flow. To enhance sensitivity perform lab, combines several biosensing (1) carbon nanotube (CNT) primary antibody immobilization, (2) sensitive ECL (electrochemiluminescence) detection, (3) charge-coupled device (CCD) detector measuring light signal generated ECL. Using sandwich ELISA assay, detected SEB at concentrations as low 0.1 ng ml(-1), which is similar reported conventional ELISA. fluidics can be operated syringe does not require power operation. This point-of-care (POC) useful carrying various assays other complex medical
