Melatonin reduces early changes in intramitochondrial cardiolipin during apoptosis in U937 cell line.

摘要: Cardiolipin (CL) is found exclusively in the inner mitochondrial membrane. CL deficiency leads to an alteration stability of membranes, increased permeability as well a decreased respiratory rate, and therefore mitochondria which are completely dysfunctional. It known that reactive oxygen species (ROS) cause decrease variation content, concomitantly formation transition pore facilitates release cytochrome c (cyt c) into cytosol. Melatonin (Mel), secretory product pineal gland, potent efficient endogenous radical scavenger. has been shown protect, various biomolecules, such DNA, membrane lipids, cytosolic proteins from oxidative damage. To evaluate protective role Mel, we have studied U937 cells treated with UV-B irradiation. In our model, administration 1 mM Mel before irradiation showed significant protection apoptotic cell death, particular, structure function were preserved through pathways when preincubated exposure. The cardiolipin-sensitive probe 10-nonyl acridine orange (NAO) was used monitor changes lipids. Our data suggest treatment protects ROS this suggests possible link reduction phenomenon.
