摘要: The goal of this study was to identify and characterise the major allergen(s) sesame seed. Detection specific IgE proteins performed with Pharmacia CAP System Western blotting, after separation by SDS-PAGE, using sera from 28 subjects diagnosed as allergic sesame. allergen separated gel filtration chromatography identified selective proteolysis followed peptide sequence analyses, employing electrospray-ionization mass spectrometer. Twenty-four had sesame-specific IgE. A 14 kDa protein belonging 2S albumin family recognised 22 24 used. Subjecting HPLC Lys C yielded 3 peptides, but only one reacted positively in dot blot test. This peptide, corresponds whole chain residues 24-94. reactivity most indicates that is allergen, later precursor; its which test evidently contains an epitope(s). Some minor allergens, higher molecular weight, were also revealed.
