摘要: Liposomes have been widely exploited as antigen delivery systems for a variety of diseases including leishmaniasis. These vesicles can be prepared in various ways which may affect the immunogenicity encapsulated antigens. In this study we compared vaccine potentiality three cationic formulations with Leishmania donovani promastigote membrane antigens (LAg) and best vesicle was evaluated long-term protection against experimental visceral We immunized mice LAg multilamellar (MLV), dehydration–rehydration (DRV) reverse-phase evaporation (REV) challenged them parasites ten days after vaccination. MLV or DRV induced almost complete protection, while alone entrapped REV exhibited partial resistance. Protection observed incorporated predominantly Th1 evidenced by elicitation significantly high DTH, IgG2a antibodies IFN-γ. demonstrated durable cell-mediated immunity weeks vaccination could also resist challenge strongly. Field trials L. were unsatisfactory mainly due to lack an appropriate adjuvant. Cationic when used adjuvant protein sustained immunity. Adjuvant potential utilized design subunit vaccines.
