摘要: The testis-specific linker histone H1t gene is transcribed exclusively in mid to late pachytene primary spermatocytes. Tissue specific expression of the mediated large part through elements located within proximal promoter. Previous work transgenic animals showed that a unique 40 bp promoter element designated H1t/TE essential for spermatocyte-specific expression. contains GC-box, which perfect consensus binding site members Sp family transcription factors. We have shown Sp1 and Sp3 are present testis cells from 9-day-old adult rats spermatocytes early spermatids they can bind H1t/GC-box. Mutagenesis GC-box reduced activity. Furthermore, CpG dinucleotide was totally unmethylated rat where but it methylated liver silenced. These previous studies supported importance suggested factors contribute gene. In this study, we show co-transfection vectors leads an upregulation activity several cell lines including GC-2spd cells. However, very low seen grown at 39 degrees C, correlates with lower levels these elevated temperature. Upregulation by also cotransfected NIH3T3 C127I lines. On other hand, does not lead cell-cycle dependent H1d
