摘要: Gliomas, characterized by their progressively invasive phenotype, express integrin alpha3beta1 as a major receptor for the extracellular matrix both in vivo and vitro. Since has been shown to be specific laminin-5 (alpha3beta3gamma2), we examined effects of purified human on adhesion, migration invasion glioma cells. Among different types laminin variants other proteins including fibronectin vitronectin, was most potent promoting adhesion kinds Laminin-5-mediated were specifically inhibited monoclonal antibodies against alpha3 beta1 chains, confirming role receptor. Invasion reconstituted basement membrane (i.e., Matrigel) cells also selectively stimulated laminin-5. Out results show that is stimulant cell invasion. The immunohistochemical distribution gamma2 chain, subunit unique laminin-5, showed it expressed tumor parenchyma tissues. Expression alpha3, beta3 chains tissues lines demonstrated at messenger RNA level reverse transcription polymerase chain reaction. Our results, taken together, may involved phenotype malignant gliomas vitro vivo.
