New synthetic applications of sialic acid aldolase, a useful catalyst for KDO synthesis. Relation between substrate conformation and enzyme stereoselectivity

摘要: In the sialic acid aldolase-catalysed addition of pyruvate with D-arabinose leading to a mixture 3-deoxy-D-manno-oct-2-ulosonic (KDO) and 4-epi-KDO (both in pyranose furanose form), high concentration acceptor have been found increase percentage KDO up 83%. synthesis scaled enzyme membrane reactor is reported. New condensation products obtained from L-xylose, L-allose, D-altrose D-ribose as substrates are described well. Kinetic parameters for cleavage reaction between xylose arabinose given. Some apparent relation stereoselectivity conformation stereochemistry at carbon C-3 substrate discussed.
