摘要: Abstract Recent lentiviral-based microRNA (miRNA) library screening has identified miRNA-7 (miR-7) as an anti‑angiogenic miRNA in human umbilical vein endothelial cells (HUVECs). However, the underlying mechanism of miR-7 suppression angiogenesis remains largely unknown. In present study, we report that inhibition promoted by upregulating vascular growth factor (VEGF) and directly targeting Kruppel-like factor 4 (KLF4). Downregulation tube formation HUVECs, accompanied upregulation mRNA protein levels both VEGF KLF4. targeted KLF4 demonstrated luciferase reporter assay mimics decreased Furthermore, bioinformatic analysis revealed presence multiple DNA-binding elements promoter. Chromatin immunoprecipitation (ChIP) antibody specifically pulled down promoter HUVECs. ectopic overexpression induced levels. addition, silencing inhibited inhibitor Our results is a direct target transcription activator VEGF. These findings indicate miR-7-KLF4-VEGF signaling axis plays important role regulation suggesting potential agent for development anti-angiogenic therapeutics diseases solid tumors.
