摘要: The cancer stem cell (CSC) theory states that many types of cancer, including nasopharyngeal (NPC), are initiated from and maintained by CSCs, which may be responsible for tumor relapse resistance to therapy. It is imperative cells (NPCSCs) specifically targeted eradicate NPC prevent recurrence. Epigallocatechin-3-gallate (EGCG) inhibits progression attenuating NF-κB p65 activity, upregulated in CSCs plays an important role epithelial–mesenchymal transition (EMT). purpose this study confirm the self-renewal migration inhibitory effects EGCG toward NPCSCs clarify its mechanism activity. We enriched characterized collecting spheroid-derived grown serum-free medium (SFM) examined on characteristics studied underlying mechanisms using soft agar colony assays, transwell reverse transcriptase polymerase chain reaction (RT-PCR), Western blot analysis, immunofluorescence staining, xenograft studies. spheroids lines acquired CSC traits underwent EMT. inhibited NPCSCs’ reversed EMT, combined treatment with cisplatin reduced growth xenografts. Moreover, activity modulating cellular localization decreasing transcriptional regulation Twist1 expression. a novel therapeutic target NPCSCs, inhibition activated offer effective NPC.
