摘要: With the aim of identifying innovative therapeutic strategies for osteosarcoma patients who are refractory to conventional chemotherapy, we analyzed in vitro effects blockage autocrine circuits. Since insulin-like growth factor-I receptor (IGF-IR)–mediated loop is relevant osteosarcoma, activity IGF-IR-blocking antibody αIR3 both sensitive and multidrug-resistant cell lines. Only limited effects, however, were observed, suggesting simultaneous existence other Indeed, a representative panel 12 human lines, addition IGF-IR-mediated circuit, demonstrated also mediated by epidermal factor as well presence nerve factor, low-affinity tyrosine kinase A great majority osteosarcomas. Therapies based on inhibition single circuits may have only whereas use suramin, drug which, besides activities, non-selectively interferes with binding factors their receptors, appears promising alternative, drug-resistant cells.
