摘要: SUMMARY: A mutant stain, Brevibacterium 19, defective for the enzyme nitrile hydratase but retaining all amidase activity of wild-type R312, was isolated. This is genetic evidence in favour hypothesis that nitrile-hydratase due to a single enzyme, structural gene which lost strain 19. The specific activities and K m were determined 12 different substrates. The affinity increased as number hydrogen-bonding positions substrate increased, decreased with more spatial crowding hydrocarbon chain. enhanced by nucleophilic hydrophilic properties side chain substrate.
