摘要: Abstract Apolipoprotein E (apoE) is synthesized by the liver and many peripheral cells. Rat ovarian granulosa cells synthesize secrete apoE, this apoE production increased agents that increase cellular cAMP. In these studies of cell synthesis we have examined effect stimulate various kinases, including protein kinases A, G, C. The content mRNA was measured simultaneously. Cholera toxin (1.25 micrograms/ml), dibutyryl-cAMP (5 mg/ml), forskolin (10(-4) M), all which cAMP, accumulation in medium 7-10-fold. On other hand, dibutyryl-cGMP (20 mg/ml) has no on or secretion. phorbol ester 12-O-tetradecanoylphorbol 13-acetate (100 ng/ml), a kinase C stimulator, increases 8-10-fold, while 4 alpha-phorbol 12,13-didecanoate, inactive congener, such effect. cAMP maximal at 48 h, 72-96 h culture. data indicate whose effects are mediated activation A C, but not production. These amount secreted temporally preceded messenger RNA. Together, suggest regulation rat could involve transcriptional post-transcriptional mechanisms.
