摘要: article i nfo Article history: Methylphenidate (MPH) treatment in boys diagnosed with ADHD is reported to decrease the risk of drug abuse adulthood. Similarly, MPH appears cocaine preference male rats during conditioned place (CPP) tests. However, effects on later use girls/ women or CPP female rodents have not been fully examined, nor a clinically-relevant dose and/ administration route thoroughly studied. Here, Sprague-Dawley (n=34/sex/treatment) were treated orally 3×/day postnatal days (PNDs) 29-50 water 3 mg MPH/kg, producing serum levels within human clinical range. assessments (10 mg/kg, ip) (PNDs 62-71) indicated MPH-treated less active pre- and postconditioning sessions (pb.04), but there no significant MPH-related differences conditioning strength. Baseline open field activity at PND 84 that females more than same-sex controls (pb.05). A challenge mg/ kg, elevated similarly both sexes. As an anhedonia measure, saccharin solution intake PNDs 87-90 effects. Estrous cycle phase did appear affect response assessments. Hormonal 90 63% higher corticosterone relative (pb.05), finding deserves further investigation. These results address some major issues surrounding animal models provide additional support for lack severe long-term behavioral adolescent MPH.
