摘要: Human-induced pluripotent stem cells (hiPSCs) are considered to be potentially able differentiate into all human cell lineages and thus hold promise as an unlimited source for replacement therapies in clinical applications. Definitive endoderm (DE) formation is the first crucial step development of visceral organs such liver, lung, pancreas so forth. Therefore, efficient generation DE ensures eventual target used therapy. In present study, Matrigel-coated poly(lactic acid)/gelatin (PLA/gelatin) nanofibrous scaffolds were utilized investigate proliferation differentiation hiPSCs cells. Analyses DE-specific markers including Sox17, FoxA2, Gooscoid (Gsc) genes revealed higher levels mRNA protein expression differentiated cultured on PLA/gelatin than two-dimensional (2D) culture. Our results showed that three-dimensional (3D) cultures could significantly promote comparison with 2D Also using small molecules inducer definitive 1 (IDE1) signaling Activin A Wnt3a enhance A/Wnt3a being more potent both 3D compared IDE1. The this study may have impact tissue engineering therapy organs-related diseases.
