摘要: Glucagon response to insulin-induced hypoglycaemia is impared in diabetes, but the mechanism not established. Pancreatic A cell hyporesponsiveness adrenergic or cholinergic stimulation could contribute impairment. We therefore compared plasma glucagon responses intravenous infusion of adrenaline (1200 ng kg(-1) min(-1) for 20 min) injection agonist carbachol (50 micrograms kg(-1)) chloral hydrate-anaesthetized rats made diabetic with use streptozotocin (80 mg subcutaneously) 6 weeks before and anaesthetized control rats. Insulin was infused intravenously reduce glucose levels below 1.8 mmol L(-1). As expected, reduced by approximately 45% streptozotocin-diabetic controls (P = 0.045). During infusion, increased 277 +/- 92 pg mL(-1) 0.009) 570 137 0.002). Thus, doubled Following injection, were raised 1211 208 < 0.001) only 555 242 0.049). 58% 0.028). conclude that carbachol-stimulated secretion concomitantly rats, whereas adrenaline-induced exaggerated. suggest a pancreatic responsiveness impairment diabetes.
