摘要: We previously reported that the adoptive transfer of lymphokine-activated killer (LAK) cells plus repetitive injections recombinant interleukin 2 (IL 2) produced a marked reduction in established pulmonary metastases from variety murine sarcomas. The requirement for exogenous administration IL prompted subsequent examination role vivo function transferred LAK cells. proliferation and migration patterns lymphoid C57BL/6 mice were examined after i.v. alone, i.p. injection or combination 2. A model labeling DNA dividing was used which injected with 5-[125I]-iodo-2'-deoxyuridine (125IUdR) and, 20 hr later, their tissues removed counted gamma analyzer. index (PI) calculated by mean cpm organs experimentally treated control mice. In animals given lungs liver demonstrated little if any uptake 125IUdR above saline-treated controls (PI = 2.5 0.8, respectively, on day 5), whereas same receiving 6000 U alone displayed higher radiolabel incorporation 7.1 5.9, respectively). When 2, showed an additional increase uptake. spleen, kidneys, mesenteric lymph nodes, treatment (6000 U) elevated PI values not, however, additionally increased also administered. To separate stimulatory effects host lymphocyte similar cells, these studies repeated immunosuppressed 500 rad total body irradiation. Pre-irradiation sufficiently reduced endogenous expansion stimulated so as to allow demonstration induced confirmed actively proliferating under influence Substitution "normal" fresh cultured (without splenocytes, irradiated did not result tissues. Histologic corroborated findings assays revealed extensive 2.(ABSTRACT TRUNCATED AT 400 WORDS)
