摘要: Mantle cell lymphoma (MCL) is a clinicopathologic entity that difficult to diagnose on histopathologic criteria. Approximately 50% 70% of MCL contain t(11;14)(q13;q32) translocation involving the cyclin D1 gene. Irrespective this rearrangement, almost all show overexpression gene at mRNA level. Other B-cell non-Hodgkin's lymphomas (NHL) do not rearrangement or D1. We developed an immunohistochemical assay detect protein conventional formalin-fixed, paraffin-embedded biopsies using well-defined monoclonal antibody DCS-6. Expression in tumor cells was compared with expression endothelial and fibroblasts. An exclusively nuclear staining pattern observed. Moreover, directly observed by immunoblot analysis same antibody, as well occurrence genomic rearrangements within BCL-1 locus. Of 13 were analyzed immunohistochemistry immunoblot, 12 showed both techniques, whereas no 39 other NHL. additional studied either 11 also overexpression. Two morphologically indistinguishable from but aberrant immunophenotype (CD5 negative, CD10 positive) lacked These results underscore significance specific marker for MCL. Detection tissues DCS-6 can be applied routine diagnostic purposes.
