摘要: The aim of this study was to investigate the effects calcium channel blockers on tertbutyl hydroperoxide (TBH) induced liver injury using isolated perfused rat hepatocytes. Rat hepatocytes were immobilized in agarose threads and with Williams E medium. Hepatocyte by addition (1 mM) perfusion medium 30 min after either verapamil or diltiazim. observed monitoring functional metabolic competence ultrastructural morphological examination Verapamil (0.5 reduced lactate dehydrogenase leakage TBH-injured as compared controls (154+/-11% vs. 247+/-30%). Lipid peroxides production pretreatment oxygen consumption increased verapamil. protein synthesis activity at both levels granular endoplasmic reticulum free polysomes cytoplasm decreased ATPase activity. Diltiazem qualitatively effective It is concluded that hepatocyte oxidative injury, exhibited hepatoprotective properties. effect accompanied a decrease activity, which may implicate normalization Ca2+i TBH intoxication.
