摘要: Hypocrea jecorina, the sexual teleomorph of Trichoderma reesei, has long been favored as an industrial cellulase producer, first utilizing its native system and later augmented by introduction heterologous enzymatic activities or improved variants enzymes. Expression proteins in H. jecorina was once considered difficult when target variant a cellulase. Developments over past nearly 30 years have produced strains, vectors, selection mechanisms that continued to simplify streamline protein expression this fungus. More recent developments fungal molecular biology pointed way toward fundamental transformation ease efficiency important host. Here, 1) we provide historical perspective on advances biology, 2) outline host strain engineering, transformation, selection, strategies, 3) detail potential pitfalls working with organism, 4) consolidated examples successful outcomes from our laboratory.
