摘要: We have developed a quick and low-cost genomic DNA extraction protocol from yeast cells for PCR-based applications. This method does not require any enzymes, hazardous chemicals, or extreme temperatures, is especially powerful simultaneous analysis of large number samples. can be efficiently extracted different species (Kluyveromyces lactis, Hansenula polymorpha, Schizosaccharomyces pombe, Candida albicans, Pichia pastoris, Saccharomyces cerevisiae). The involves lysis colonies liquid culture in lithium acetate (LiOAc)–SDS solution subsequent precipitation with ethanol. Approximately 100 nanograms total 1 × 107 cells. by this suitable variety applications (including colony PCR, real-time qPCR, sequencing) amplification fragments ≤3500 bp.
