摘要: The development of in vitro methods for regulatory embryotoxicity testing is challenging since the understanding chemical effects on mammalian still poor. aim project to identify marker genes during cell differentiation murine embryonic stem cells, order predict specific target tissues. present study focusing expression pattern by using semi-quantitative reverse transcripatse (RT)-PCR key involved cardiomyocytes development; i.e. Oct-4, Brachyury, Nkx2.5 and alpha myosin heavy chain (α-MHC). Two reference chemicals with well-known vivo data have been analysed this approach: retinoic acid lithium chloride. Retinoic has selected as a teratogenic affecting several tissues, whereas chloride described affect cardiovascular system. We demonstrate that already affects early stage germ layer formation, which was demonstrated change Oct-4 Brachyury gene expression. As we expected, cardiac (Nkx2.5, α-MHC) also modified. In contrary not changed treatment. case, observed modification normal pattern, α-MHC Nkx2.5, demonstrating later heart development. These suggest inclusion selective organ an established test allows drugs
