Mycobacterium tuberculosis infection increases the number of osteoclasts and inhibits osteoclast apoptosis by regulating TNF-α-mediated osteoclast autophagy.

摘要: Osteoarticular tuberculosis, a chronic inflammatory disease characterized by Mycobacterium tuberculosis (M.tb) infection, has become serious problem in China. The present study was conducted to determine the mechanism of action tumor necrosis factor (TNF)-α pathogenesis osteoarticular tuberculosis. number osteoclasts tissue samples detected tartrate-resistant acid phosphatase staining. Autophagy and apoptosis were western blotting, reverse transcription-quantitative PCR, transmission electron microscopy TUNEL results showed that autophagy increased lesions patients with compared osteoarthritis samples. Moreover, activation osteoclast inhibited infected M.tb, expression level TNF-α. TNF-α enhanced autophagic activity M.tb-infected cell apoptosis. These findings indicated M.tb infection induced production regulating TNF-α-mediated autophagy, revealing new for
