Characterization of a xenograft model of human ovarian carcinoma which produces intraperitoneal carcinomatosis and metastases in mice

摘要: A new xenograft model for human epithelial ovarian carcinoma, with extensive intraperitoneal (i.p.) carcinomatosis as the predominant disease manifestation, is described. Cells from established NIH:OVCAR-5 cell line were injected i.p. into 6- to 8-week-old Swiss nude mice. Comparative analyses between cells cultured in vitro and tumor derived ex vivo performed assess histologic features, immunohistochemical markers, hormonal receptor expression, adhesion extracellular matrix molecules chromosomal constitution. Macroscopically, extent of development appeared be site-dependent survival was dose-dependent. Advanced characterized by solid burden ascites parenchymal invasion, lymphatic metastases vascular dissemination. Individual nodules exhibited developing neovasculature absence mature basement membrane. Despite some loss cellular differentiation advanced disease, antigenic expression preserved, distinguishing these origin. Karyotyping demonstrated multiple numeric structural abnormalities. Serum CA 125 levels consistently elevated only tumor-bearing This murine closely resembles aggressive process which efficacy systemic therapeutic modalities can investigated.
