EGFR antisense oligonucleotides encapsulated with nanoparticles decrease EGFR, MAPK1 and STAT5 expression in a human colon cancer cell line.

摘要: Epidermal growth factor receptor (EGFR) is over-expressed in several human cancers. This would suggest that inhibition of EGFR a reasonable approach for cancer treatment. In this study we investigated blocking and its effects on the mediated signaling such as MAPK STATb HT29 cells. For aim used FITC-labeled antisense oligonucleotides encapsulated with PAMAM nanoparticles to inhibit expression. Cellular uptake was by fluorescence microscopy flow cytometry analysis. The effect expression cells examined real time PCR Western blots, which showed decreased level mRNA protein. addition, results confirmed had an effective role reduction dependent downstream genes. conclusion, down regulated EGFR-mediated
