摘要: The objective of this study was to determine if human oviduct specific glycoprotein (huOGP) would associate with hamster ovarian oocytes and sperm during in vitro incubation. huOGP used these studies partially purified from hydrosalpinx fluid. Hamster samples were incubated culture medium without huOGP. Association assessed by indirect immunofluorescence assay using a polyclonal antibody prepared against Intense fluorescence the zona pellucida, bright but uneven perivitelline space, observed following incubation presence A similar more uniform pattern when oviductal (positive controls) alone. Fluorescence absent assayed preimmune serum. association pellucida space appeared be since thyroglobulin, large molecular weight glycoprotein, serum albumin, major protein fluid, did not nor inhibit included medium. antiserum However, fluoresced uterine CUPED, which had previously been shown bind cat Sperm also brightly as positive control. Solubilization membrane proteins postincubation analysis 1-D SDS-PAGE followed immunoblotting failed show an sperm. Electron microscopy both pre- postsolubilization confirmed that membranes removed process. In conclusion, suggests may vivo, whereas true for vivo. serve facilitate process fertilization early embryonic development within oviduct. © 1994 Wiley-Liss, Inc.
