摘要: Plasmodium circumsporozoite protein (CSP) is a major surface antigen present in the sporozoite (Spz) stage of malaria parasite. RTS, S vaccine, most clinically advanced consists large portion falciparum CSP (PfCSP). A highly infectious, recombinant rodent malaria, yoelii parasite bearing full-length PfCSP, PfCSP/Py Spz, was needed as tool to evaluate role PfCSP mediating, protective, anti-malaria immunity mouse model. transgenic parasite, generated by inserting construct expressing at locus P. gene double cross-over homologous recombination. Then biological and protective properties Spz were determined. This produced up 30,000 mosquito salivary glands, which equal or even higher than number wild-type parasites. Five bites PfCSP/Py-infected mosquitoes could induce blood infection BALB/c mice. The current study has demonstrated successful establishment clone that able express Importantly, this can be infectious both vector mouse, mammalian host. new expresses may become useful for researchers investigate against
