Cell Behavior within Nanogrooved Sandwich Culture Systems

摘要: Grooved topography and inherent cell contact guidance has shown promising results regarding proliferation, morphology, lineage-specific differentiation. Yet these approaches are limited to 2D applications. Sandwich-culture conditions developed bridge the gap between 3D culture, enabling both ventral dorsal surface stimulation. The effect of grooved is accessed on orientation elongation in a highly controlled manner, with simultaneous independent stimuli two sides. Nanogrooved non-nanogrooved substrates assembled into quasi-3D systems variable relative orientations. A plethora sandwich-culture created by seeding cells lower, upper, or substrates. Software image analysis demonstrates that F-actin acquires substrate which initially seeded, independently from second top substrate. Contrasting morphologies observed, higher for nanogrooved than conditions. Correlated an increased pFAK activity vinculin staining conditions, point enhanced stimulation versus control pivotal role initial cell-biomaterial cellular alignment highlighted, providing important insights tissue engineering strategies aiming guide response through mechanotransduction approaches.
