Ketamine and thiopental sodium: individual and combined neuroprotective effects on cortical cultures exposed to NMDA or nitric oxide

摘要: Background An N -methyl-d-aspartate (NMDA) blocker, ketamine, has been shown to be neuroprotective both in vivo and vitro . However, ketamine is not commonly recommended for use patients suffering from cerebral ischaemia because of its adverse neurological effects. We hypothesized that combined administration thiopental sodium (TPS) would highly effective protecting cortical neurones ischaemia, with possibly reduced dosages. Methods examined the degree neuroprotection provided by various concentrations TPS, alone combination, cultures exposed NMDA or a nitric oxide-releasing compound (NOC-5) 24 h. The survival rate (SR) E16 Wistar rat was evaluated using photomicrographs before after exposure these compounds. Results SRs 30 μM NOC-5 were 15.0 (3.8)%, 12.8 (3.1)%, respectively. Higher doses (5, 10 50 μM) but lower ( P Only highest dose TPS (50 improved NOC exposure. This influenced ketamine. Conclusions These data indicate low, clinically relevant offer significant during prolonged NOC. Combinations exhibited enhanced against NMDA-induced neurotoxicity. Hence, combinations two common i.v. anaesthetics agents could developed protect brain ischaemia.
