摘要: AIM: To establish a convenient immunoassay method based on recombinant antigen preS1(21-119 aa) to detect anti-preS1 antibodies and evaluate the clinical significance of in hepatitis B. METHODS: The expression plasmid pET-28a-preS1 was constructed, large quantity fragment HBsAg protein obtained. preS1 purified by Ni2+-IDA affinity chromatography used as coated indirect ELISA streptavidin-biotin system for detection sera from HBV-infected patients. For follow-up study, serial were collected during course 21 patients antibodies, antigen, HBV-DNA other serological HBV markers analyzed. RESULTS: highly expressed soluble form E. coli (30 mg•L⁻¹), easily high purity over 90% one step Ni2+-IDA-sepharose 6B chromatography. antigenicity determined 150 g•L⁻¹ SDS-PAGE, Western blot direct ELISA. Recombinant successfully applied which could sensitively serum specimens acute or chronic B Results showed that more than half 19 produced recovery disease, however, response only found few In study 11 with positive profile, clearance occurred 6 10 5-6 mo, seroconversion HBeAg disappearance 1 treated lavumidine, antiviral agent. CONCLUSION: high-purity prepared gene Using this conveniently detective established. Preliminarily trial occurrence suggests short-term time, means health improvement disease.
