摘要: OBJECTIVE To investigate the feasibility of differentiation human induced pluripotent stem cells (iPSCs) into epidermal-like cells. METHODS (1) Human strain iPSCs were plated on-to trophoblast inactivated Fb mouse embryos and cultured in complete medium embryonic cells, subcultured by collagenase IV digestion method. The morphology growth observed under inverted phase contrast microscope, stained with alkaline phosphatase (AKP). incomplete to observe ability embryoid body formation. (2) inoculated onto 6-well plate covered amniotic membrane culture as induction group. Other on without control Morphological changes two groups observed. Expressions integrin beta1 CK19 determined immunocytochemical staining. RESULTS showed a typical cell clone-like clear boundary, they proliferated vigorously These AKP-positive. formed trophoblast-free suspension conditions. After 4 days co-culture, clones surface group, part positive. Most died, no positive found CONCLUSIONS can be differentiated induction, it lays an experimental basis for providing new source seed skin tissue engineering.
