Equine epidermis: a source of epithelial-like stem/progenitor cells with in vitro and in vivo regenerative capacities.

摘要: Besides the presence of somatic stem cells in hair follicles and dermis, epidermis also contains a subpopulation cells, reflecting its high regenerative capacity. However, only limited information concerning epidermis-derived epithelial-like stem/progenitor (EpSCs) is available to date. Nonetheless, this cell type could prove itself useful skin reconstitution after injury. After harvesting from equine epidermis, purified were characterized as EpSCs by means positive expression for CD29, CD44, CD49f, CD90, Casein Kinase 2β, p63, Ki67, low cytokeratin (CK)14 negative CD105, CK18, Wide CK, Pan CK. Furthermore, their self-renewal capacity was assessed adhesion well suspension. Moreover, isolated differentiated toward keratinocytes adipocytes. To assess capacities EpSCs, six full-thickness wounds made: three treated with platelet-rich-plasma (EpSC/PRP-treated), while remaining administered carrier fluid alone (PRP-treated). The dermis EpSC/PRP-treated significantly thinner exhibited more restricted granulation tissue than did PRP-treated wounds. increases vascularization, elastin content, follicle-like structures. In addition, combining PRP treatment enhanced repair clinical application.
