摘要: Abstract Recent experimental studies have demonstrated that membrane systems can be used to separate protein variants differ by only a single amino acid residue, although the selectivity and purification factor were relatively low. The objective of this study was examine theoretically effects surface charge density pore size distribution on separation high performance tangential flow filtration. Model calculations based partitioning charged sphere into cylindrical pore, including shift in pH within pores due preferential positive hydrogen ions, are good agreement with literature data for myoglobin succinylated variant. Significantly greater obtained reducing mean increasing density, both which enhance electrostatic interactions arising from small difference between native Purification factor–yield plots developed evaluate performance. Proper selection properties gave 100-fold at 90% product yield differing lysine group.
