摘要: Abstract Nonporous particles of microsize were prepared by the dispersion polymerization styrene and glycidyl methacrylate chemically modified to introduce amino groups on surface grafting with either hexamethylenediamine or N-methyl-1,3-propanediamine. Aminated then coupled phosphorylated single-stranded polynucleotides at 5′-end through covalent linkages. The affinity columns packed these polynucleotide-immobilized effectively retained DNA, which could base-pair immobilized sequence. Bound DNAs be eluted yield a sharp peak using an aqueous solution 0.4 M NaOH. nonspecific adsorption due electrostatic interaction between polynucleotide residual particle via amination was significant only reduced high salt (NaCl) concentration. A higher concentration in elution result portion complementary nonretained fraction. However, insignificant column DNA-immobilized effective for microanalysis sequence-specific DNA.
