摘要: AIM: To determine the optimal concentration for inducing differentiation of human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) into neuron-like cells, although it is understood that all-trans retinoic acid (ATRA) regulates cell proliferation in nervous system by modulating balance between mitosis and apoptosis. METHODS: The abilities ATRA to promote apoptosis as well neural were assessed cultured hUC-MSCs morphological observation, MTT assay, annexin V-FITC/PI flow cytometry immunocytochemistry. RESULTS: data showed low concentrations (0.5 µmol, 0.25 µmol) had no effect on number cells. However, treatment with 1.0 µmol or 2.0 induced a 24.16% 52.67% reduction number, respectively, compared vehicle-treated cultures. Further, 4.0 potent almost adherent recovered after 24h. We further 0.5 caused these express characteristic markers neuronal progenitor CONCLUSION: Taken together, we conclude has dose-dependent influence hUC-MSCs. These findings have implications use ATRA-differentiated study degeneration diseases.
