摘要: Abstract Multiple bleb formation on cell membrane is common during death. The effects of α-tocopherol and glutathione (GSH) tert -butyl hydroperoxide (TBH)-induced changes in rat hepatocytes were studied. Over 60 min exposure, TBH (0.5–2.0 m ) caused a dose-dependent blebbing. Cells pretreated with buthionine sulfoximine, GSH synthesis inhibitor, had significantly greater blebbing lactate dehydrogenase (LDH) leakage under 0.5 treatment as compared to cells without pretreatment. However, the protective effect disappeared when concentration was increased 2.0 . In presence α-tocopheryl succinate (TS) pretreatment, it noted that formation, expressed percentage bearing blebs, average size, or onset blebbing, partially suppressed even TBH-induced thiobarbituric acid reactive substances LDH completely abolished by TS Accompanying membrane-insoluble actin decrease immunoblot assay also TS. These results indicated intracellular status are important abnormality. Furthermore, plays defensive role against exhausted TBH.
